Sermorelin

Sermorelin is associated with binding to the growth-hormone-releasing hormone receptor (GHRHR), a class B G-protein-coupled receptor expressed on anterior pituitary somatotrophs. Because it reproduces the active fragment of endogenous GHRH, research suggests it engages the same recognition site rather than acting on the somatostatin or ghrelin/GHS receptor systems. In study models, this receptor selectivity is thought to underlie its endogenous, secretagogue-style profile.

Upon GHRHR engagement, research suggests coupling to Gs proteins that activate adenylate cyclase, raising intracellular cyclic AMP and downstream protein kinase A activity. This signaling cascade is associated in study models with increased transcription and pulsatile secretion of stored growth hormone from somatotroph cells. Elevated PKA tone has also been linked to calcium influx that supports the exocytotic release step.

GHRH-family peptides are described in research as components of the hypothalamic-pituitary neuroendocrine axis, where the native ligand originates from arcuate nucleus neurons. Sermorelin itself is studied primarily for its peripheral pituitary action, and limited direct neural activity beyond this neuroendocrine signaling is described in the research. Some study models additionally associate GH/IGF-1 axis tone with sleep-architecture and slow-wave sleep parameters.

By promoting pituitary-driven GH pulses, Sermorelin is associated in research with downstream hepatic IGF-1 generation and the broader somatotropic regulatory axis governing protein, lipid, and glucose metabolism. Because the response remains under hypothalamic somatostatin feedback, study models suggest it tends to preserve physiologic pulsatility rather than overriding the loop. Researchers also explore its relationship to body-composition, recovery, and metabolic regulatory pathways.
