Kisspeptin

Kisspeptin-10 is the active C-terminal fragment associated with binding the KISS1 receptor (KISS1R, also known as GPR54), a G-protein-coupled receptor expressed densely on GnRH neurons of the hypothalamus. Research suggests this receptor engagement is the principal upstream event that the broader reproductive endocrine cascade depends on. In study models, the decapeptide retains much of the binding affinity of the longer parent peptide while showing a comparatively short circulating half-life.

Upon receptor occupancy, KISS1R is described in the research as coupling to Gq/11 proteins, activating phospholipase C and driving inositol-trisphosphate and diacylglycerol production with subsequent intracellular calcium mobilization. These second-messenger events are associated in study models with depolarization of GnRH neurons and downstream ERK/MAPK pathway engagement. The net intracellular effect investigated is a stimulus that promotes pulsatile neuropeptide release rather than a sustained tonic signal.

Beyond its hypothalamic role, kisspeptin signaling has been studied in limbic and olfactory-associated brain regions, where research explores correlations with sexual and emotional processing. Neuroimaging studies in humans have associated kisspeptin administration with modulated activity in networks linked to attraction, reward, and mood. The peptide is therefore described in the literature as having central as well as endocrine relevance, though the precise neural mechanisms remain an active area of investigation.

Systemically, kisspeptin sits upstream of the hypothalamic-pituitary-gonadal axis and is associated in study models with influencing luteinizing hormone and follicle-stimulating hormone release, and in turn downstream sex-steroid signaling. Research also explores KISS1 expression outside the brain, including in reproductive and metabolic tissues, suggesting broader regulatory involvement. Reported effects on autonomic and metabolic parameters are preliminary and remain under study.
